To add insult to injury, Jesus was flagged offside. City thought they had drawn level midway through the half when Aymeric Laporte drilled home a sidefooted shot from 10 yards but it was correctly ruled out by VAR and on-field referee Mike Dean when replays showed Jesus had controlled the ball with the help of his arm in the build-up.
But it was yet more frustration for De Bruyne, who could not stop the goal nor initiate a response from his City team-mates. The only blemish on the evening for Spurs was a suspected groin injury suffered by Toby Alderweireld in the latter stages.
It was a danger sign for Spurs, though. City seemed to have recovered their poise. City had most of the ball. But they did not present the Spurs defence with enough problems.
Too many of their forwards seem to have lost their mojo. Bernardo Silva and Riyad Mahrez always wanted one touch too many. Jesus is a fine player but he is not as ruthless as Sergio Aguero.
Who is? Sometimes, De Bruyne looks exasperated with what he sees in front of him. City tried to rid themselves of their ponderousness and De Bruyne shot just wide five minutes after half time.
An attack broke down, Spurs played the ball out of defence to Harry Kane, who was in his own half. He turned and ran at the City defence, which was suddenly hopelessly stretched.
Kane waited and timed his pass to Lo Celso to perfection. Lo Celso took a touch and drilled his shot past Ederson.
Spurs had managed two shots on target and scored two goals. By the time Sterling and Phil Foden came on soon afterwards, it was too late and City find themselves marooned in mid-table.
Guardiola has signed his new contract. Now he has to start earning it. Mike Dean blows his whistle for full time and what a huge result that is for Tottenham, who have earned a much-deserved win over City to firmly establish themselves as title contenders.
City were dominant in the first half and enjoyed the lion's share of possession, but their pretty football did not equate to goals, and Spurs demonstrated the sort of ruthlessness in defence and attack of champions.
It was Son who opened the lead for the hosts, before second half sub Giovanni Lo Celso put the result beyond doubt.
Mourinho will be absolutely delighted with that performance, while Guardiola has plenty to ponder as his side lie 8 points adrift of the new league leaders with a game in hand.
City have a chance to get the ball in the box. Chance City! No Spurs supporters would have expected to dominate possession against Pep Guardiola's Manchester City but everything else that they would have hoped for from their team was delivered.
Tottenham were organised and they were diligent in their duties. When they could attack, they did so with genuine quality. That quality has been enough to carry them to the top of the Premier League table and there is a growing belief that in this most unusual of seasons they might just be able to stay there.
This was not them at their most expansive but those qualities have also been apparent in scoring 21 goals this season. Tottenham now have quality on the bench too.
For Guardiola, his concerns are on the pitch. A new contract but the same old problems for the Manchester City boss, ones that have not been solved by his new centre-back pairing.
The defensive line continues to be breached as opponents sit deep and pick them off on the counter-attack. Lessons from the Leicester defeat have not been learned.
